Silica Gel Column Chromatography


Chromatography is a technique used to isolate the various components of a mixture and this application is very important in analysis of bio-molecules. The technique is used to separate and analyze the complex DNA sequences and other compounds, and also the concentration of the samples. Chromatography is a separation process involving two phases, stationary and mobile phase. Mixture to be analyzed is adsorbed in stationary phase and mobile phase is passed into it, eventually compounds of mixture get separated on basis of rate of adsorption and solubility. In the process of column chromatography, a glass tube is filled with adsorbent, that is either alumina or silica gel up to a level of one thirds of the length of the glass tube. Then, it is soaked with selective solvent. Sometimes, column is filled with slurry, an amalgam of adsorbent and solvent. The column should not have any space, should be well packed. In this method, less polar compound will be eluted first. Because, less polar compound will be less adsorbed in polar stationary phase. Eventually, more polar compound will come out.
